A Training Course on Cell Culture and Molecular Diagnostic Techniques 

15th July to 4th August, 2010

Objective:

The process of culturing of animal cells outside the body from which they have been obtained is known as animal cell culture (ACC). To carryout the process successfully, all the conditions should mimic the in vivo environment. The ACC is carried out under strict laboratory conditions of asepsis, sterility and controlled environment involving temperature, gases and pressure. Production of therapeutically significant biological compounds like hormones and proteins on an industrial scale has been made simpler, faster and more efficient by the use of animal cell lines instead of the living animal themselves.

ACC can be employed to mass produce the virus. Passively, ACC can be employed to reduce the virulence of particular virus strains by cultivating them on cells other than target cells which the virus infects followed by repeated passaging. In present scenario, livestock productivity is greatly hampered by various infectious and communicable diseases, so proper diagnosis has immense importance as far as their treatment and control is concerned. Commercial kits are very expensive and not affordable in the country due to poor economic status of the livestock rearing farmers. Therefore, it is important that highly important diagnostic tests should be developed indigenously based on the Indian strains of pathogens. In the past years, DNA based diagnostic tests have become popular due to their sensitivity and specificity in certain countries. Therefore, the present practical training course is designed to provide hands on training on basic molecular techniques used for diagnosis of pathogens in blood, semen, clinical tissues, faecal samples and characterization of pathogens and breeds of animals. The training programme will also include theoretical aspects of molecular biology and recombinant DNA tools.

Course contents:
  • Basic requirements for animal cell culture laboratory
  • Primary and continuous cell culture
  • Media and reagents preparation for animal cell culture
  • Storage and revival of animal cells
  • Propagation of animal virus in cell line
  • Extraction and purification of nucleic acid of viral agents from various sources like blood, semen, clinical tissues, faecal samples, etc..
  • Electrophoretic separation of nucleic acid and protein
  • RNA-Polyacrylamide gel electrophoresis for segmented viruses.
  • Synthesis of cDNA and RT-PCR based amplification of viral genes.
  • DNA markers like RAPD,RFLP etc.
  • Molecular cloning of viral genes in suitable vectors
  • Confermation of clones by colony touch
  • Sequence alignment tools e.g. BLAST
  • Analysis of nucleotide sequence.
  • Phylogenetic analysis of sequence data.
Course Fees: Indian participants are requested to pay a sum of Rs. 5000/- (Rs five thousand only) while for foreign delegates US$ 200 per week as registration fee.

The registration fee shall be deposited in cash at the time of registration on 15th July, 2010.

Laboratory and computing facilities: The molecular diagnostic laboratories are well equipped with modern equipments and other lab wares.

Accommodation: Arrangements for the stay of the participants during the training program will be made in faculty house of the university on the payment basis.

Number of participants: The maximum number of participants shall not exceed 20.

Participants and eligibility: Participants are invited from ICAR Institutes/ SAU/Basic Science Institutes/ Sate Governments/Private Organizations. They should be graduates having interest in the training.

How to apply: The application for participation may be sent in prescribed format, duly forwarded by Head of the institution. It should reach to the Course Director latest by 1st July, 2010 by post, fax or email. TA & DA of the participants will be borne by participants/sponsoring institutions/ organisations etc. The participants will also have to pay for their boarding and lodging charges during the training program.

The organizers of the course will not bear any expenses on account of the participants. The participants will be informed about their selection by 7th July, 2010.
