Project Title: Translational Decoding of TCR-Neoantigen Dynamics in Oral Precancerous Lesions
Progression towards Frank Cancer: Integrating Spatial Sequencing and AI for Early Prediction
Biomarker Development.
Oral Squamous Cell Carcinoma (OSCC) remains one of the leading causes of cancer-related mortality in
India, largely driven by late-stage diagnosis and high recurrence rates. While oral precancerous lesions,
such as Leukoplakia, are frequently identified during routine screenings, the lack of definitive molecular
biomarkers makes it nearly impossible to predict which lesions will undergo malignant transformation. This
project aims to bridge that gap by shifting the focus toward early intervention and immunogenomic risk
stratification.
The project involves -
(i) combining high-throughput TCR sequencing with existing somatic
mutation, gene expression, and methylation data from oral cancer patients.
(ii) utilizing cutting-edge spatial
technologies to understand the regulatory roles and architectural nuances of the immune milieu in
independent patient cohorts.
(III) developing a predictive AI/ML algorithm that integrates immunogenomic
features for early disease progression risk assessment.
